MacConkey Agar is a selective and differential culture medium designed for isolating gram-negative enteric bacteria and differentiating lactose fermenters from non-fermenters. It contains bile salts and crystal violet to inhibit the growth of gram-positive organisms, and lactose as the fermentable carbohydrate. Neutral red is used as a pH indicator, turning pink in acidic conditions resulting from lactose fermentation. Lactose fermenters, such as Escherichia coli, produce pink colonies, while non-fermenters, like Salmonella and Shigella, form colorless or pale colonies. MacConkey Agar is extensively used in clinical and environmental microbiology to identify and differentiate enteric bacteria based on their lactose fermentation ability.
