MacConkey Agar is a selective and differential culture medium designed for isolating gram-negative enteric bacteria and differentiating lactose fermenters from non-fermenters. It contains bile salts and crystal violet to inhibit the growth of gram-positive organisms, and lactose as the fermentable carbohydrate. Neutral red is used as a pH indicator, turning pink in acidic conditions resulting from lactose fermentation. Lactose fermenters, such as Escherichia coli, produce pink colonies, while non-fermenters, like Salmonella and Shigella, form colorless or pale colonies.
